What is PEG-MGF?

PEG-MGF (PEGylated Mechano Growth Factor) is a polyethylene- glycol-conjugated form of the 24-residue C-terminal peptide of IGF-1Ec — a splice variant of insulin-like growth factor 1 produced in skeletal muscle in response to mechanical loading. The MGF peptide drives satellite-cell activation and muscle- fibre repair; PEGylation extends the otherwise-short plasma half-life of the bare peptide.

Mechanism of action

Mechano Growth Factor is the C-terminal Ec-exon peptide produced by alternative splicing of the IGF-1 gene under mechanical stress. It activates a distinct receptor pathway from full IGF-1 and drives muscle satellite-cell proliferation and recruitment to repair sites. PEGylation extends plasma half-life from minutes to approximately 72 hours, supporting sustained signalling in experimental protocols.

Research context

Investigated in preclinical models of skeletal-muscle injury repair, muscle hypertrophy in response to mechanical loading, and age-related sarcopenia. The PEGylated form is preferred over bare MGF for protocols requiring sustained exposure.
