Bacteriostatic water: what it is and why it is used

Bacteriostatic water appears alongside almost every lyophilised peptide listing, usually without explanation. It is a specific, well-defined preparation, and the property that distinguishes it has a narrow purpose: it allows a container to be entered more than once. This guide explains the composition, the difference from plain sterile water, and the handling consequences.
Composition
Bacteriostatic water is sterile water containing approximately 0.9% benzyl alcohol as a preservative. That is the entire difference from sterile water. Benzyl alcohol is bacteriostatic rather than bactericidal — it inhibits the multiplication of bacteria rather than killing an established population.
The distinction matters. The preservative does not sterilise a contaminated solution and does not compensate for poor technique. It suppresses growth of organisms introduced in small numbers during ordinary handling.
Why the preservative exists at all
Plain sterile water is sterile only until the container is entered. Once a needle passes through the septum, the contents are no longer guaranteed sterile, which is why sterile water for injection is packaged as single-use.
Bacteriostatic water is designed for multi-dose containers: the preservative allows the same vial to be entered repeatedly over a period without organisms introduced at each entry establishing a population. In a laboratory context that means one vial can serve to reconstitute several samples over days or weeks rather than being discarded after one use.
What it does not do
Three common misconceptions:
- It does not stabilise the peptide chemically. Benzyl alcohol addresses microbial growth. It has no effect on oxidation, deamidation, hydrolysis or aggregation — the routes covered in storage and stability.
- It does not extend chemical shelf life. A reconstituted solution still degrades on the same chemical timetable.
- It does not rescue contaminated material. Bacteriostatic is not the same as bactericidal.
Compatibility considerations
Benzyl alcohol is not inert with respect to every application. Two considerations recur in laboratory work:
- Cell-based assays. Benzyl alcohol has measurable effects on membrane fluidity and is cytotoxic above certain concentrations. Where a reconstituted stock is diluted into cell culture, the carried-over preservative concentration should be calculated rather than assumed negligible, and a vehicle control run.
- Analytical work. Benzyl alcohol absorbs in the UV and will appear on a chromatogram. If a reconstituted sample is being re-analysed by HPLC, the preservative peak needs to be accounted for rather than integrated as an impurity.
Where either applies, plain sterile water or an appropriate buffer, single-use, is usually the better choice.
Choosing between the two
A simple decision rule covers most cases:
- Single use, immediate consumption, or a system sensitive to benzyl alcohol — sterile water or the buffer specified for the assay.
- A stock that will be entered several times over a period — bacteriostatic water.
Neither choice substitutes for the solubility requirements of the specific sequence. Standard practice for a multi-use vial: swab the septum before each entry, use a fresh needle each time, avoid introducing air, and store between uses at the temperature appropriate to the dissolved peptide rather than to the water.
