What is Kisspeptin-10?
Kisspeptin-10 (KP-10, Metastin 45-54) is a 10-residue fragment of kisspeptin (Metastin) — a 54-residue peptide encoded by the KISS1 gene that is a master regulator of hypothalamic GnRH neurons and consequently of the reproductive endocrine axis. The 10-residue C-terminal fragment retains full KISS1R (the kisspeptin receptor) binding activity.
CAS 374683-28-0, molecular formula C₆₃H₈₃N₁₅O₁₃S, molecular weight 1302.55 g/mol. ChEMBL ID CHEMBL3422406.
Mechanism of action
Kisspeptin-10 binds the KISS1R (also called GPR54) receptor on GnRH neurons in the hypothalamus, triggering GnRH release and consequently downstream LH and FSH secretion from the anterior pituitary. The kisspeptin/KISS1R/GnRH axis is a principal regulator of reproductive function across mammals; loss-of-function mutations in KISS1 or KISS1R produce hypogonadotropic hypogonadism in humans.
Research context
Investigated in clinical and preclinical studies of reproductive-axis function, hypogonadotropic hypogonadism, and as a diagnostic probe for hypothalamic GnRH-neuron function. Kisspeptin-10 has also been investigated in infertility-related clinical research.
Analytical specifications
Every batch of Kisspeptin-10 supplied by NMChem is characterised by reversed-phase HPLC for purity determination and by mass spectrometry for identity confirmation. Certificate of Analysis (COA) documents are issued per batch and made available on request.
Identity is confirmed by electrospray-ionisation mass spectrometry (ESI-MS); the observed mass falls within ±2 Da of the theoretical monoisotopic mass calculated from the residue sequence. Purity is determined by reversed-phase HPLC on a C18 column with a trifluoroacetic-acid-modified water/acetonitrile gradient and reported as the area-under-curve percentage of the main peak at 214 nm. NMChem specification is ≥99% main peak. Material ships as lyophilised powder and must be reconstituted in sterile water or bacteriostatic water immediately before use.
UK regulatory status
Supplied as a research-grade reference standard for laboratory use only. Not a licensed medicinal product in the United Kingdom and not approved by the MHRA for human or veterinary administration. Sale is restricted to researchers, institutions and laboratory professionals; the compound must be retained within research premises. End-user compliance with the Human Medicines Regulations 2012, the Misuse of Drugs Act 1971 (where applicable) and local institutional biosafety policy is the responsibility of the buyer.
