What is CJC-1295 (with DAC)?
CJC-1295 with DAC is the long-acting variant of the CJC-1295 family — a GHRH (1-29) analogue carrying a Drug Affinity Complex (DAC) at the C-terminus. The DAC is a maleimidopropionic-acid moiety that forms an irreversible thioether bond with cysteine-34 of circulating human serum albumin, anchoring the peptide to albumin and extending its plasma half-life from minutes to approximately 6–8 days.
CAS 863288-34-0 (shared with the No-DAC form in some databases), molecular formula C₁₆₅H₂₆₉N₄₇O₄₆, molecular weight 3647.28 g/mol. Wikidata QID Q5012018.
Mechanism of action
The mechanism of GHRH-receptor activation is identical to CJC-1295 (No DAC); the difference is pharmacokinetic. The DAC moiety provides covalent albumin-anchoring that extends plasma residence to approximately 6–8 days, producing a much smoother and more sustained GH-pulse profile compared to the rapid pulsatile dynamics seen with the No-DAC form.
Research context
Investigated specifically in protocols that require sustained GH-release modulation rather than acute pulses — typically rodent models of body-composition change and bone-density adaptation studies over weeks to months. The extended half- life is the principal reason for selecting CJC-1295 (with DAC) over the No-DAC variant.
Analytical specifications
Every batch of CJC-1295 (with DAC) supplied by NMChem is characterised by reversed-phase HPLC for purity determination and by mass spectrometry for identity confirmation. Certificate of Analysis (COA) documents are issued per batch and made available on request.
Identity is confirmed by electrospray-ionisation mass spectrometry (ESI-MS); the observed mass falls within ±2 Da of the theoretical monoisotopic mass calculated from the residue sequence. Purity is determined by reversed-phase HPLC on a C18 column with a trifluoroacetic-acid-modified water/acetonitrile gradient and reported as the area-under-curve percentage of the main peak at 214 nm. NMChem specification is ≥99% main peak. Material ships as lyophilised powder and must be reconstituted in sterile water or bacteriostatic water immediately before use.
UK regulatory status
Supplied as a research-grade reference standard for laboratory use only. Not a licensed medicinal product in the United Kingdom and not approved by the MHRA for human or veterinary administration. Sale is restricted to researchers, institutions and laboratory professionals; the compound must be retained within research premises. End-user compliance with the Human Medicines Regulations 2012, the Misuse of Drugs Act 1971 (where applicable) and local institutional biosafety policy is the responsibility of the buyer.
